Antioxidative Effects of Traditional Korean Herbal Medicines on AAPH - induced Oxidative Damage

Abstract
Herbal extracts from traditional Korean prescriptions, commonly used for anti-aging, were investigated for in vitro and in vivo antioxidative potentials. Antioxidative potentials were evaluated based on DPPH (1,1-diphenyl-2-picrylhydrazyl) radical quenching capacity, inhibition of pyrogallol auto-oxidation, and peroxyl radical scavenging activity. Yungmijihwang-Won (YJ), Yollyung-gobon-Dan (YG), and Palmi-Hwan (PM) showed relatively high scavenging activities of free radicals. Antioxidative potentials of these extracts were also investigated through in vivo experiment. In the mouse brain and liver tissue treated with AAPH [(2,2¢¥-azobis(2-amidinopropane) dihydrochloride], YJ decreased the thiobarbuturic acid reactive substance (TBARS) production and the protein degradation level, which are used as indices of oxidative stress, and increased total antioxidant status and glutathione. However, activities of antioxidative enzymes such as SOD, catalase, and glutatione peroxidase, were not changed by these extracts. From these results, it could be assumed that water-soluble extracts of YJ, YG, and PM have antioxidative potentials and, thereby, protect cells against oxidative stress. However, increased antioxidative potentials of the extract-treated mice did not appear to be related to antioxidative enzyme activity in vivo but were considered to be related to antioxidative potentials of herbal compounds of the prescriptions.
